Xenobiotic Metabolic Enzymes Bioactivation and Antioxidant Defense

Xenobiotics are commonly referred to as xenobiotics that humans digest or inhale and are not produced by the body, such as foods, drugs, other chemicals, and air pollutants. Metabolic conversion of xenobiotics to facilitate their removal from the body is catalyzed by activation and detoxification enzymes. These enzymes are essential for bioactivation and detoxification of xenobiotics. Many xenobiotics are non-toxic in nature; however, some of them become cytotoxic or carcinogenic after metabolic conversion. Reactive intermediates or metabolites generated during metabolic conversion are generally electrophilic in nature and are either cations or neutral compounds, including reactive oxygen species and free radicals. Metabolic reactive intermediate-mediated oxidative stress can cause damages to cellular components such as impairment of protein functions, membrane lipid peroxidation, and formation of DNA adduct.

Toll-like Receptors in Inflammation

Toll-like receptors (TLRs) are critical mediators of the innate immune response in mammals. This family of receptors recognizes a variety of microbial products or motifs and initiates the host response to infection. Examples include TLR4 which recognizes lipopolysaccharide (LPS) from Gram-negative bacteria, TLR3 which recognizes viral double-stranded RNA, and TLR9 which recognizes CpG DNA motifs, found in both viruses and bacteria. All TLRs possess an intracellular region termed the Toll-IL-1 receptor-Resistance (TIR) domain which is essential for signaling from these receptors. The specificity of signaling from individual TLRs arises from differential utilization of adapter proteins. This specificity results in a tailoring of the host defense response depending on the microbe being sensed. TLRs are increasingly being implicated in both infectious and inflammatory diseases, notable examples being sepsis, inflammatory bowel disease, atherosclerosis, and asthma. There is, therefore, great interest in targeting TLRs therapeutically since a disruption of TLR function will result in a decrease in the production of inflammatory mediators.

The first extended study of Rowe’s defense of atheism on the basis of evil, Nick Trakakis comprehensively assesses the large body of literature that has developed in response to Rowe’s work, paying particular attention to two strategies employed by critics: firstly, the appeal to mystery – the idea that God may well have reasons for permitting evil that lie beyond our comprehension; and secondly, the appeal to theodicies, where this involves offering explanations as to why God allows evil to abound in his creation (free will theodicies, for example, argue that God could not prevent us from acting wrongly without at the same time curtailing or removing our free will). Trakakis unearths significant difficulties in both strategies, and concludes that – absent any evidence in support of theism – the God of theism must be judged to be "beyond belief".
